Puyol missed the first game and Gerard Pique was booked and therefore suspended for the second leg as well in the 2-1 Arsenal victory two weeks ago.
Puyol suffered a reccurence of the knee injury which kept him out of the first leg, and Barcelona will be forced to rely on Gabriel Milito and Eric Abidal on Tuesday.
